When the bacterial suspensions are placed in the 42 water bath, the outside environment of the cell is becomes slightly higher in temperature than the inside of the cell. This unbalance in temperature creates a draft in which the plasmids are sweeped into the bacterial cells through the pores of the cell membrane.
k) It is important that the bacterial suspensions are cooled on ice for 15 minutes in a calcium chloride environment because this allows the bacterial cells to recover from the heat shock and stay alive...
