Aim:
I will be culturing samples of known bacteria using standard procedures. The bacteria I will be culturing is Escherichia-coli (e-coli). After incubation I will note the appearance of growing colonies. This will give me experience in aseptic techniques. The appearance of the agar plates will indicate if I have been successful in excluding contamination by other micro-organisms.
Time:
I will need about 1 hour in the laboratory to set up the agar plates and, 48 hours later, about 30 minutes to examine them.
Equipment and Materials:
Sterile equipment: 1 petri dish containing nutrient agar; 2 dropper pipettes
Sterile water
Overnight broth cultures of micro-organism (Escherichia coli)
Inoculation loop
Glass spreader and foil covered beaker of 70% industrial methylated spirit for flame sterilisation
Bunsen burner
Discard pot containing disinfectant
Marker pen
Tape to secure Petri dishes
Incubator at 25-30 C
Safety and risk assessment:
Wear protective clothing and eye protection.
Do not open the Petri dishes after incubation. (BIOHAZARD)
